PRINCETON, NJ, USA I October 22, 2018 I Bristol-Myers Squibb Company (NYSE:BMY) today announced the results of a new analysis from the Phase 3 CheckMate -214 study, demonstrating that therapy with Opdivo (nivolumab) plus Yervoy (ipilimumab) in patients with previously untreated advanced or metastatic renal cell carcinoma (RCC) was associated with significantly longer treatment-free survival (TFS).

With a minimum follow-up of 30 months, 36% of patients who received Opdivo plus Yervoy were still alive and not requiring subsequent therapy vs. 16% of patients who received sunitinib. At two years in patients who discontinued therapy, 19% of Opdivo plus Yervoy patients vs. 6% of sunitinib patients remained treatment free. Data from the analysis (Presentation #874P) will be featured in a poster presentation at the European Society for Medical Oncology (ESMO) 2018 Congress in Munich, Germany, October 19-23.

“The results from this analysis of CheckMate -214 provide important insights into the potential for sustained clinical benefit with the combination of nivolumab and ipilimumab in patients with advanced renal cell carcinoma, for whom there is a considerable unmet need,” said CheckMate -214 investigator David F. McDermott, MD, Director of the Immuno-Oncology Program at Beth Israel Deaconess Medical Center.

TFS was significantly longer with Opdivo plus Yervoy vs. sunitinib regardless of best overall response (p<0.0001). Two years after discontinuation, 42% of patients treated with the combination vs. 12% on sunitinib with complete or partial responses remained free of subsequent treatment; at the same time point, 12% of combination-treated patients and 6% of sunitinib patients with stable disease remained treatment free. Similarly, TFS benefit of Opdivo plus Yervoy vs. sunitinib was also observed across PD-L1 subgroups. For patients with PD-L1 ≥1%, 27% of patients from the Opdivo plus Yervoy arm remained treatment free at two years after discontinuation compared to 8% from the sunitinib arm (p=0.0002). For patients with PD-L1 <1%, 18% of patients from the Opdivo plus Yervoy arm vs. 5% from the sunitinib arm remained treatment free two years after discontinuation (p<0.0001).

About CheckMate -214

CheckMate -214 is a Phase 3, randomized, open-label study evaluating the combination of Opdivo plus Yervoy vs. sunitinib in patients with previously untreated advanced or metastatic renal cell carcinoma (RCC). Patients in the combination group received Opdivo 3 mg/kg plus Yervoy 1 mg/kg every three weeks for four doses followed by Opdivo 3 mg/kg every two weeks. Patients in the comparator group received sunitinib 50 mg once daily for four weeks, followed by two weeks off before continuation of treatment. Patients were treated until progression or unacceptable toxic effects. The primary endpoints of the trial are overall survival (OS), progression-free survival (PFS), and objective response rate (ORR) in an intermediate to poor-risk patient population (approximately 75% of patients). The majority of alpha was allocated to OS. Safety is a secondary endpoint.

Overall, at the time of the primary study analysis, 320 (75%) patients in the Opdivo plus Yervoy group and 359 (85%) patients in the sunitinib group had discontinued treatment, most commonly due to disease progression (42% and 58%, respectively) or study drug-related adverse events (23% and 11%, respectively). In patients who discontinued, treatment-free survival (TFS) was significantly longer with Opdivo plus Yervoy than sunitinib (p<0.0001); 18 months after discontinuation, 19% vs. 4% of patients remained treatment-free, respectively.

OPDIVO® (nivolumab) is indicated for the treatment of patients with metastatic non-small cell lung cancer (NSCLC) with progression on or after platinum-based chemotherapy. Patients with EGFR or ALK genomic tumor aberrations should have disease progression on FDA-approved therapy for these aberrations prior to receiving OPDIVO.

OPDIVO® (nivolumab) is indicated for the treatment of patients with metastatic small cell lung cancer (SCLC) with progression after platinum-based chemotherapy and at least one other line of therapy. OPDIVO® (nivolumab) is indicated for the treatment of patients with locally advanced or metastatic urothelial carcinoma who have disease progression during or following platinum-containing chemotherapy or have disease progression within 12 months of neoadjuvant or adjuvant treatment with platinum-containing chemotherapy.
